CIDRを抜くと同時にPGF2αを投与すべきなのか、抜く前日にPGF2αを投与するべきなのかについて少し考える機会がありましたので、自分の調べられる限りでまとめてみました。
牛の排卵同期化(Synchronization of ovulation)・定時人工授精(Timed artificial insemination:TAI)プログラムには様々な方法がありますが、二つに大別されます。
1つ目は、オブシンク法を代表とする、GnRH処置により存在する主席卵胞を排卵させ、新しい卵胞波を誘導する方法です。
2つ目は、膣内留置型プロジェステロン製剤(CIDR)と安息香酸エストラジオール(Estradiol benzoate:EB)処置により、FSHとLHの分泌を抑制し、卵巣に存在する主席卵胞を含む卵胞群を閉鎖させ、新しい卵胞波を誘導する方法です。
そして、これらプログラムにおいて、高い受胎率を得るための戦略として、次の5つの要素が重要視されています1)。
- GnRHやCIDR + EBにより、確実に新しい卵胞波を誘導すること。
- 誘導された新しい卵胞波の卵胞が発育する間、黄体期レベルの血中プロジェステロン(P4)濃度を維持すること。
- PGF2αを用いて、黄体を完全に退行させることにより、P4濃度を限りなく低下させ、TAI時のエストラジオール(E2)濃度を上昇させ発情を誘起すること。
- 適切なサイズに成熟した卵胞をGnRHやE2を用いて確実に排卵させ、授精と排卵を確実に同期させること。
- 排卵後に形成される黄体が適切に機能し、高く安定したP4濃度が維持されること。
今回のテーマは、3番目の要素について関係があると思いますので、その点に絞って調べてみました。
多くの研究で、TAI時のP4濃度と受胎率の関係が報告されています。TAI時においてP4濃度が十分に低下することは、精子の輸送、卵子の質、子宮内膜や胚の遺伝子発現に有利な環境を与え、受胎性を向上させます2)。しかしながら、P4濃度が十分に低下せずにTAIが行われているケースが現状のTAIプログラムでは存在し3-9)、その原因として不十分な黄体退行が指摘されています8, 10, 11)。
不十分な黄体退行の原因は、
①PGF2α処置からTAIまでの間隔が短いこと
②間隔が確保されても完全な黄体退行に至らないこと
の2つが考えられています12-14)。
それゆえ、この不十分な黄体退行の問題を克服するためのPGF2α処置について、様々な方法が検証されています。
今日のテーマは、①に該当すると考えます。①に対する対策として、PGF2α投与およびCIDR抜去からTAIまでの時間を確保しTAI時のP4濃度をできる限り低下させる方法が行われています。CIDR開始日をday 0とした場合、CIDRを8日間留置し、抜去時の前日(day 7)にPGF2αの処置を行う場合と抜去当日(day 8)にPGF2αの処置を行うプログラム(図1)において受胎率を比較した結果、day 7投与群の方がday 8投与群よりもday 10のTAI時におけるP4濃度が低い牛の割合が多く、受胎率が高かったです5)(表1)。
また同文献では、PGF2α処置日に関係なくTAI時のP4濃度が低い牛において受胎率が高いことも報告されています。さらに、PGF2α投与からTAIまでの日数が3日よりも4日の方が発情発現率が高く、受胎率には差がないものの、妊娠60日までにおける妊娠損失率が低いという受胎性の向上がみられました。
TAI時の発情発現は受胎性にとって重要で、発情が発現した牛の方が受胎率が高く15-18)、受胎後の妊娠損失率5, 19)も減少することが報告されています。発情が強く長く持続する牛は、発情が弱く短い牛よりも、発情時のP4濃度が低く、E2濃度が高く推移し、受胎率が高い結果でした20)。P4濃度だけでなく、TAI時のE2濃度も受胎率と正の相関があります21)。TAI前のGnRH投与時にP4濃度が高く、またはE2濃度が低い牛は、LHサージと排卵率が低下します22-25)。よって、PGF2α投与からTAIまでの期間を1日長く設けることは、排卵前のE2濃度の上昇26)、発情発現率の増加5)、排卵率の向上27)、排卵後の黄体期における妊娠に必要な適切なP4濃度の維持を可能とし26, 27)、受胎性を向上させる5, 27)ことにつながると考えられます。
しかしながら、TAI時のP4濃度と受胎性について、牛の品種による差があることも報告されています。乳用牛ではTAI時のわずかなP4濃度の上昇は受胎率を低下させますが、肉用牛においてはPGF2α投与66~72時間後、つまりPGF2α投与後3日のTAI時におけるP4濃度は受胎率に影響を与えなかったことが報告されています8)。肉用牛は乳用牛よりもTAI時のP4濃度がより低い牛の割合が高く、P4によるLHサージの抑制を受けにくいため、より高い排卵率が期待でき、受胎率低下が起きにくいことが理由として考えられています8)。このように、肉用牛は乳用牛ほどのTAI時のP4濃度による受胎性への影響は受けにくいと考えられているものの、CIDR抜去と同時にPGF2αを投与し、その後1日にTAIを実施した場合に関しては、TAIを2日後に実施した場合に比べて排卵率が低下し受胎率が低下することが報告されています27)。
まとめますと、CIDR抜去前1日にPGF2αを投与し、PGF2αからTAIまでの間隔をより長く設定することによって、より完全な黄体退行を誘導し、P4濃度が可能な限り低下した状態でTAIを行うことは、受胎性にとって重要な要素であり、特に乳用牛では肉用牛に比べその影響をより受けるものと考えられます。しかしながら、低いP4濃度の状態を少し長く取り過ぎて、その中で卵胞が発育すると卵子がエイジング(受胎能力ピークを過ぎてしまう)を起こしてしまいますのでその点には注意が必要になります。以下が少し参考になると思われる記事です。
