牛の飼養管理や栄養状態を改善し、自然発情がしっかり来て、それによる人工授精という流れが最も大事であることは確かだと自分は思います。しかし、発情が来ない・妊娠しない牛に対する薬剤を使った繁殖管理が、現時点ではまだ重要な役割を果たしていることも事実ではあります。
発情同期化は繁殖管理法の一つであり、様々な種類がありますが、日本や南米ではエストロジェン製剤が使用できるため(北米や欧州では使用禁止)、それを併用したプログラムがあります。
安息香酸エストラジオール(EB)とCIDR(例:イージーブリード)、またはPRID(EBのカプセル付き)を用いたプログラムは汎用性が高く比較的安価であるため、広く利用されています。特に、乳牛よりも肉用牛での使用が多いのではないかと思います。
今回は、CIDRとEBを併用したプログラムの理論について、自分が後輩に説明できる程度にまとめてみようと思います。
まずCIDRを腟内に留置することによって牛を高P4濃度に暴露させることで始まり、P4濃度の上昇により、黄体形成ホルモン(LH)のパルス状分泌が抑制されると報告されています(1, 2)。
LHのパルス状分泌はGnRHの拍動性分泌と連動しており(3)、主に主席卵胞の選抜と主席卵胞の発育、成熟に関与するホルモンであるため(4)、LHが低下するとその時点で存在する主席卵胞が閉鎖退行します(5, 6)。
次に、CIDR挿入と同時にEBを投与します。CIDR挿入によりP4濃度を上昇させた状態においてEBを投与することで血液中のE2濃度を上昇させると、負のフィードバック作用により視床下部でのGnRHの分泌が抑制され、下垂体前葉での卵胞刺激ホルモン(FSH)分泌が一時的に低下します(7)。
投与したEBが肝臓で代謝され血液中のE2濃度が低下すると、低下したFSHの分泌が24~48時間後に再度上昇を開始し(8, 9, 10)、FSHの上昇後約1日で新規卵胞波の出現が起きると報告されています(11)。
FSHは卵胞の発育に関与するホルモンであり、卵胞波発現に関わるため(12)、EB投与後約3~5日で直径3~4 mm程度の卵胞が発育を開始し、新しい卵胞波が出現するといわれています(8, 13, 14, 15)。
卵胞波として動員された卵胞はFSHやLHの作用を受け、黒毛和種繁殖雌牛の場合1日当たり1.5~2 mmの速度で排卵に向けて発育を続けます(16)。
卵胞波の発現から3日程経過すると、卵胞波の中の最大卵胞の顆粒層細胞にLH受胎体が発現し、E2産生が行われるようになります(17)。
E2は下垂体前葉に作用しFSHの分泌を抑制する(18)ため、卵胞波の卵胞群の中で主席卵胞として選抜されなかった次席以下の卵胞は発育が維持できず閉鎖していきます。
しかしながら、最大卵胞はLH受容体を発現することで、その発育の主役となる性腺刺激ホルモンはFSH依存性からLH依存性にシフトするため、LHパルスの刺激を受けて閉鎖することなく発育が続きます(19, 20)。
選抜された卵胞を主席卵胞といい(21)、選抜時における最大卵胞の平均直径は8.5 mmであり,次席卵胞の平均直径は7.7 mmとされています(22)。
黒毛和種繁殖雌牛における選抜時の最大卵胞の平均直径は8.9 mmであり、次席卵胞は6.8 mmと報告されています(16)。
主席卵胞の発育の過程でCIDRを除去し、同時にPGF2αを投与することでその時点で存在する黄体を退行させ、よりP4濃度を低下させます。
黄体が退行し、血中P4濃度が低下することでLHのパルス状分泌が亢進し(1, 2),主席卵胞がさらに発育を続け、成熟卵胞で産生されたE2が分泌され、血中E2濃度が上昇します(23)。
その後、EB製剤を投与し血中E2濃度をさらに上昇させることで、視床下部からGnRHサージが誘導され、連動して下垂体前葉からLHサージが起こり(24)、成熟卵胞がEB投与後42~60時間で排卵します(25, 26)。
そのため、EB投与後24~36時間で授精を行うのが一般的です。
また、血中E2濃度が上昇するにつて発情強度が増加することが報告されています(27)。
発情同期化または定時人工授精プログラムを適用した場合、人工授精時に発情がある方で受胎率が高く(28,29)、さらに発情の有無に加えその発情の強度の増加による受胎率の上昇(30)と、強度に加え排卵率の上昇による受胎率の向上(31)が報告されているため、CIDR除去後にEBを排卵誘起剤として投与することによってE2濃度を上昇させることは受胎性にとって有用であるとされています。
以上が、これまでの報告を参考にまとめさせていただいた、CIDRと安息香酸エストラジオールを用いた繁殖プログラムの理論になります。
